MCA-Gly-Asp-Ala-Glu-pTyr-Ala-Ala-Lys(DNP)-Arg-NH2 acts as a substrate for many protein tyrosine phosphatases. After dephosphorylation, the substrate is treated with chymotrypsin which cleaves the Tyr-Ala bond resulting in an increase in fluorescence (Ex 325 nm, Em at 393 nm).
