Calpain Inhibitor III is a potent, cell-permeable inhibitor of calpain I and II (Ki = 8 nM) in addition to cathepsin B (Ki = 24 nM). Calpains are calcium-dependent cysteine proteases involved in a wide range of physiological processes. Calpain inhibitors can protect against necrotic neuronal death and, to a lesser extent, apoptotic death. Calpain inhibitors are useful tools for studying diseases, especially those involving neurodegeneration. Calpain Inhibitor III has been shown to inhibit neurofilament degradation in neurodegenerative diseases.
