BHD-C2C2-PipZ is an ionizable lipid which can form PEG-free lipid nanoparticles (LNPs) for RNA delivery. PEG-related immunogenicity is a well-known problem, but substituting tripolyphosphate resulted in stable LNPs that demonstrated increased expression compared with traditional LNPs. Hepatic tolerability was comparable to traditional LNPs and cytokine profiles indicated no inflammation suggesting repeat dosing was possible without PEG-related side effects.
