Ac-Arg-Gly-Phe-Phe-Pro-AMC is a fluorogenic substrate suitable for the assay of cathepsin D. Cathepsin D cleaves the Phe-Phe bond and the resulting RP-AMC dipeptide can be cleaved by dipeptidyl peptidase IV (DPP IV) resulting in a fluorescent signal. Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
Cathepsin D is a lysosomal aspartyl protease whose role is to degrade and activate intracellular proteins including hormones, enzyme precursors, and growth factors. Over expression in tumors stimulates metastasis and considered a marker of poor prognosis in breast cancer.
