Z-Phe-Tyr-CHO is a reversible peptidyl aldehyde inhibitor of Cathepsin L (IC50=0.85 nM). It is selective vs cathepsin B and calpain II (IC50s=85.1 and 184 nM respectively). Cathepsin L is a lysosomal cysteine protease with an important role in initiating protein degradation. Cathepsin L is also upregulated in several cancers and has been suggested as a drug target. In addition, extracellular secretion of Cathepsin L by tumors implicates it as a potential serum biomarker for cancer progression.
