In this presentation, Dr. Petrey explored how the extracellular matrix component hyaluronan (HA) and platelets interact to drive inflammation and microvascular thrombosis in disease. Under normal conditions, a glycan-rich HA matrix coats the vascular endothelium and limits adhesion of immune cells and platelets. However, during inflammation or tissue damage, HA becomes modified into pro-inflammatory, adhesive structure capable of recruiting leukocytes and platelets to the vessel wall. Dysregulation of HA metabolism by platelets can promote excessive leukocyte recruitment, enhance inflammation, and promote microthrombi formation, contributing to chronic inflammatory pathologies like inflammatory bowel disease and other systemic inflammatory states. The talk highlighted how platelets sense the state of the HA matrix, a critical interface between inflammation and coagulation and novel therapeutic target.
