—The sphingosine kinase inhibitor screen assay is ideal as a high throughput screen in a 384-well format and is designed to pair with the BellBrook TranscreenŽ ADP2 FI Assay. The reaction starts with the addition of ATP into the sphingosine-sphingosine kinase mixture containing the screening compound. After incubation, ADP (product) is detected by the Transcreen ADP detection mixture. The amount of ADP produced is proportional to the SPHK reaction product, S1P (See Figure 1). The assay was validated using the sphingosine kinase inhibitor, 2-(p-hydroxyanilino)-4-(p-chlorophenyl)thiazole (Echelon Cat# B-0024), with sphingosine kinase 1 (Echelon Cat# E-K068) and sphingosine kinase 2 (Echelon Cat# E-K069).
