HDAC Assay Developer is an essential reagent for signal generation in HDAC or Sirtuin assays using a fluorogenic acetylated peptide substrate for HDACs based on the residues of 379-382 of p53 (Arg-His-Lys-Lys (Ac)), as site of regulatory acetylation by the p300 and CBP acetyltransferase (lysines 381, 382). An appropriate fluorogenic substrate, comprising an acetylated lysine side chain, is first incubated with a sample containing HDAC activity (nuclear or cellular extract, purified enzyme, bead-bound immunocomplex, etc.). Deacetylation of the substrate sensitizes it so that, in a second step, treatment with the HDAC Assay Developer produces a fluorophore. Typically, an ¿inhibitor stop¿ will be included in the HDAC Assay Developer (e.g. Trichostatin A for class I and II HDACs, nicotinamide for sirtuins).
Storage:
—Store product frozen at or below -70 ºC for long term storage. Freeze in working aliquots to avoid repeated thawing and freezing. Concentrate is stable for at least 3 freeze-thaw cycles, but care should be taken that both freezing and thawing steps are done quickly (rapid finger-rubbing for thawing and flash freezing in liquid N2).
