—The ratio of SM/PC is thought to be an independent risk factor for coronary heart disease. For your convenience we have combined the analysis of both phospholipids into one simple assay kit.

| Product Background: |
—In this enzyme coupled assay, phosphatidylcholine is cleaved by phospholipase D to produce choline and phosphatidic acid. Choline is oxidized by choline oxidase to betaine producing hydrogen peroxide. DAOS plus 4-Aminoantipyrine (4 AAP) in the presence of hydrogen peroxide and peroxidase results in the oxidative coupling of DAOS and 4 AAP to form a blue chromogen that is measured by absorbance at 595 nm1. Separately, sphingomyelin is cleaved into phosphorylcholine and n-acylsphingosine using Sphingomyelinase. Alkaline Phosphatase hydrolyzes phosphorylcholine and choline is oxidized by choline oxidase to hydrogen peroxide. DAOS plus 4-Aminoantipyrine (4 AAP) in the presence of hydrogen peroxide and peroxidase results in the oxidative coupling of DAOS and 4 AAP to form a blue chromogen that is measured by absorbance at 595 nm1.
—Experiments have shown the Phosphatidylcholine/Sphingomyelin Assay Kit can detect phosphatidylcholine and sphingomyelin at concentrations of 17 ug/mL using a reaction time of 4 hours.
—The kit is designed to quantify both PC and SM of 16 samples run in duplicates.
