Description:
—Echelon's Collagen IV ELISA Kit is a full-sandwich ELISA. The plate, included in the kit, has been coated with a Collagen IV capture reagent and blocked to reduce non-specific binding. The user simply adds the provided standard curve and samples to the plate and, following an incubation and plate wash, adds the detection reagent labeled with HRP. After an additional incubation and plate wash, TMB substrate is added to the plate and the colorometric reaction stopped by the addition of 1N sulfuric acid. The absorbance at 450 nm is measured and the concentration of samples determined by comparison to the standard curve.

Product Background:
—Collagen IV is a major component of the extracellular matrix and vessel basement membranes. It has been demonstrated that a variety of diseases cause alterations in Collagen IV synthesis, degradation, and release at the tissue level. Collagen IV serum levels are altered in many diseases, including liver fibrosis (with hyaluronan), autoimmune diseases (SLE, RA, etc), and vessel disruption. Collagen IV has also been implicated in metastatic cancer. Research has shown that an alteration in Collagen IV binding affects the metastasis of cancer cells. Serum Collagen IV may be indicative of Collagen IV degradation in the tissue and may correlate with metastasis. The quantitative measurement of Collagen IV may reflect the severity of the disease and assist in the monitoring of disease progression and treatment effect.
