PIP-ArraysTM are useful in determining the relative degree of protein binding to phosphoinositides.
PIP Arrays are 4 cm by 4 cm nitrocellulose membranes that have each of the eight synthetic phosphatidylinositol phosphates (PtdInsPns) spotted and dried on them, each in seven different concentrations. Each strip has 100, 50, 25, 12.5, 6.25, 3.13, and 1.58 pmol of the following phosphoinositides: Phosphatidylinositol, syn: PtdIns, PI Phosphatidylinositol (3) phosphate, syn: PtdIns(3)P, PI(3)P Phosphatidylinositol (4) phosphate syn: PtdIns(4)P, PI(4)P Phosphatidylinositol (5) phosphate syn: PtdIns(5)P, PI(5)P Phosphatidylinositol (3,4) bisphosphate, syn: PtdIns (3,4)P, PI(3,4)P2 Phosphatidylinositol (3,5) phosphate, syn: PtdIns(3,5)P, PI(3,5)P2 Phosphatidylinositol (4,5) bisphosphate, syn: PtdIns(4,5)P, PI(4,5)P2 Phosphatidylinositol (3,4,5) trisphosphate, syn: PtdIns(3,4,5)P, PI(3,4,5)P3 The phosphoinositides are spotted onto nitrocellulose membrane, for a total of 56 spots.
